New Standard Operating Procedure: Measurement and Analysis of Mitochondrial Function In Situ in Permeabilized Muscle Fibers
We are excited to announce the release of our latest Standard Operating Procedure (SOP) titled “Measurement and Analysis of Mitochondrial Function In Situ in Permeabilized Muscle Fibers”, developed by our Basic Science Trainee Commitee members Dr. Sean Ng and Magda Lensinski. The SOP aims to provide a comprehensive, step-by-step protocol using the Oroboros O2k high-resolution respirometry system to precisely measure and analyze cellular oxygen consumption, mitochondrial function, and bioenergetic efficiency, particularly in dystrophic skeletal muscle.
This new resource provides detailed methods covering permeabilized muscle fiber bundle preparation, chamber calibration, experimental titrations (pyruvate, malate, ADP titration, glutamate, cytochrome c, and succinate), and data analysis using DatLab software. Researchers can use this protocol to assess mitochondrial respiration, oxidative phosphorylation, and metabolic flexibility in health and disease states.
